Rome to Florence? Where to stay for a night, enroute?

My wife and I are planning to drive from Rome to Florence. We have 1 night which we want to spend in the Tuscany region enroute to Florence from Rome.

After reading several posts around, we are more confused. Which is the best place that we should be stopping at to spend the night. We leave from Rome after breakfast and intend to reach Florence by the next day evening. So we have time to see around along the route. Unfortunately, we have just 1 night :(

Which would be the most suitable destination for us.
We like it all - Nature, architecture, food and wine.

Val d'Orcia for sure!

The ideal location would be the Val d'Orcia area in southern Tuscany -

but anywhere between Montepulciano, Pienza, San Quirico and Montalcino would be perfect. You can drive up after breakfast, spend the morning exploring Montepulciano, have lunch there and then head on to your hotel or B&B (generally check in is early afternoon). Spend the rest of the day exploring any or all of the other towns above. The next morning after check-out you can visit anything else you missed and then head up to visit Siena for the rest of the day before heading on to Florence......... Siena really deserves more time, so if you can return another day, you can skip it and just drive up leisurely up the SS2 (Via Cassia from the Val d'Orcia, pass through Siena and then head up the Via Chiantigiana SP 222 through Chianti. You can stop in Castellina for lunch, then head up past Panzano and Greve to reach Florence by evening. Then return to Siena on another day ;-)
