Tuscany / Florence During Easter

Hello,
I am looking into planning a trip for next April; 3 days in Florence and roughly 5 days in the Tuscany region. This is a trip for me and my daughter, who will be 17 when we travel. We love art, history, would like to experience the countryside as well. No real need to tour many wineries. However, we would be arriving in the area the Wednesday before Easter, which has us there before, during and after the holiday. We are Catholic, so it would be wonderful to experience this holiday there ... but also, now am wondering if many museums would be closed during this time?
Lastly, we would stay in Florence for 3 nights ... is there a central place in Tuscany that we should consider staying and then doing day trips to say Sienna, Lucca and Val d'Orcia ... or should we do one-nighters?
